Spermatogenesis - sperm diagnostics

3 important questions on Spermatogenesis - sperm diagnostics

The predictive parameters of sperm for sperm diagnostics?

  • motility
  • volume and the concentration van the sperm
    • the darkest yellowish colour -> higher concentration of spermatozoa
  • morphology

How to calculated the concentration of sperm with the Burker counter chamber?

  1. You count the sperm cells in 100 squares of the Burker counter chamber.
    1. focus on sperm heads
    2. Count two borders of a square, it does not matter which sides you do not count
  2. 1 square is 1/4000 mm^3
  3. calculation for the concentration of sperm cells: 100 square spermcells X dulitionfactor  X 4000/100 (we did this X 1000 to get from mm3 to ml)
    1. 159 x 200 x 4000/100 x 1000 -> 1272x10^6 million spermcells/ ml

How do you evaluate the morphology of the sperm?

For morphology you make a smear and stain the sperm cells  -> Red colour enter the dead spermatozoa --> they are membrane permeable, membrane is damages.
