What is the JC-1 assay?

JC-1 is a cationic (positief geladen), lipophilic dye that selectively accumulates in mitochondria depending on their membrane potential.


It exists in two forms:
  • Monomeric form: Emits green fluorescence (~530 nm)
  • Aggregated form: Emits red fluorescence (~590 nm)

The potential is decreased when mitochondrials are damaged (less negative). Therefore less aggregation.

What will you measure when there is mitochondrial stress?

You produce more lactate. => glucose wordt niet omgezet in pyruvate maar in lactaat. Dit kan namelijk zonder zuurstof.

To which cellular mechanisms does the seahorse look?

Why do you still need rotenone/antimycin when you already add an product that causes proton leaks?

=>  FCCP geeft je maximale capaciteit, maar niet het nulniveau van mitochondriale OCR
  • => Zonder rotenone/antimycin weet je niet welke OCR nog steeds mitochondriaal is en welke van andere enzymen komt.
  • => Voor precieze berekening van reserve capacity, ATP-linked OCR en proton leak heb je dat laatste plateau nodig.
